BDNF抗体封闭对大鼠脑缺血损伤肺组织IL-6表达的影响

Effects of BDNF Antibody Block on IL-6 Expression in Lung of Rats Subjected to Brain Ischemia

  • 摘要: 目的 探讨脑缺血肺损伤后脑源性神经营养因子(BDNF)是否能调节损伤肺组织炎症因子白细胞介素-6(IL-6)的表达。方法 将大鼠随机分为假手术组、脑缺血肺损伤组、脑缺血肺损伤给予BDNF抗体封闭组(BDNF抗体封闭组)。每组13只大鼠,于BDNF抗体干预后3 d处死,5只用于免疫组化,8只用于RT-PCR和Western blot,检测BDNF封闭对IL-6定位与表达量的影响。结果 免疫组化染色显示,气道上皮细胞和巨噬细胞可见IL-6表达,胞浆染色为主。RT-PCR和Western blot检测显示BDNF抗体封闭后3 d,肺组织IL-6 mRNA和蛋白表达下降,与脑缺血肺损伤组比较,差异有统计学意义(P<0.05)。 结论 BDNF抗体封闭减少脑缺血肺组织IL-6表达,提示BDNF可能通过调节IL-6参与脑缺血肺水肿。

     

    Abstract: 【Abstract】 Objective To explore effect of brain-derived neurotrophic factor (BDNF) on IL-6 expression in the lung of rats with brain ischemia. Methods Inflammatory lung injury was induced by brain ischemia in rats that were devided into sham operation group, brain ischemia lung injury (BILI) group and brain ischemia with BDNF administration group. Lungs were harvested from rats in each group in 3 days after brain ischemia respectively. Immunohistochemistry, RT-PCR and Western blot were employed to detect the expressional changes on the mRNA and protein of IL-6 in the lung tissues. Results Expression of IL-6 was mainly found in cytoplasm of lung epithelial cell and macrophage with immunohistochemistry staining. A significant decrease of IL-6 expression was observed at 3 days after brain ischemia with BDNF antibody block. Conclusion BDNF, as a crucial neurotrophic factor, could regulate IL-6 expression in rats lung tissues with brain ischemia.
