真皮多潜能细胞注射对皮肤弹性的影响

Effect of Dermal Multipotent Cells Transplantation on Skin Elasticity in Aged Murine

  • 摘要: 目的 探讨真皮来源多潜能细胞(dermal multipotent cells,DMCs)注射对皮肤弹性的影响。方法 从3 d 龄BALB/c小鼠真皮中分离培养获得DMCs,取第3代DMCs观察形态和成脂、成骨分化。将36只12月龄BALB/c小鼠分为DMCs组(n=12)、成纤维细胞(FBs)组(n=12)及对照组(n=12),分别将10 4 个DMCs、10 4 个FBs和100 μL DMEM培养基注射入小鼠双下肢之间背部中央部位的皮下作单点注射。于2周及4周使用MPA580皮肤弹性测试仪检测小鼠皮肤弹性变化。结果 DMCs接受诱导分化后,分别成脂、成骨。接受DMCs注射的小鼠皮肤弹性较对照组在2周后有较大增加( P=0.000),但与FBs组差异无统计学意义( P=0.216)。4周后皮肤弹性不仅较对照组增加( P=0.000),且亦较FBs组增加( P=0.031)。结论 DMCs注射在小鼠皮下可以增加皮肤的弹性。

     

    Abstract: Objective To investigate the elasticity changes in aged dermis after injection of dermal multipotent cells (DMCs). Methods Dermal multipotent cells were isolated and cultured from 3-day-old BALB/c mice and then transplanted into the dermis of aged (12-week-old) BALB/c mice. Adult fibroblasts(FBs) were employed as control. At 2 and 4 weeks after the transplantation, we examined dermal elasticity by MPA580 skin test machine. Results The skin elasticity were improve at 2 weeks after the transplantion in both DMCs group and FBs group ( P= 0.000) but have no statistical difference between these two groups ( P=0.216). The different effect between these two groups appeared in 4 weeks after the transplantation ( P=0.031). Conclusion Dermal multipotent cells appear more effective than fibroblasts in increasing skin elasticity.
