寻常型银屑病TWEAK/CD163在外周血中表达的初步研究

Expression of TWEAK and Its Receptor CD163 in Peripheral Blood of Psoriasis Vulgaris (PV)

  • 摘要: 目的 初步探讨肿瘤坏死因子相关弱凋亡诱导因子(TWEAK)及CD163在寻常型银屑病(psoriasis vulgaris,PV)中的作用。方法 2016年6~12月间共收集28例PV患者和15例健康对照者外周血,ELISA法测定血清TWEAK及CD163的表达水平,real time-PCR法测定外周血TWEAK mRNA及CD163 mRNA的表达水平,流式细胞仪检测外周血中TWEAK及CD163的蛋白在单核细胞的表达比例。分析PV患者银屑病严重程度(PASI评分)与TWEAK、CD163表达的相关性。结果 纳入时28例PV均处于进展期,治疗后随访到恢复期的有10例。PV进展期血清TWEAK水平较正常对照组升高,而血清CD163水平较对照组下降,PV恢复期血清TWEAK水平较进展期下降而血清CD163水平升高;TWEAK mRNA 水平在PV低于对照组,而CD163 mRNA水平在PV和对照无明显差异;TWEAK 在单核细胞的表达率在PV进展期和恢复期均高于对照组,而CD163在单核细胞的表达率在PV进展期和恢复期均低于对照组。PV进展期和对照组单核细胞的TWEAK和CD163共表达率均高于PV恢复期(P<0.05),而PV进展期和对照组比较差异无统计学意义。血清TWEAK水平、血清CD163水平、TWEAK及CD163在单核细胞的各自表达率和共表达率均与PV严重程度无相关性(P>0.05)。结论 TWEAK/CD163炎症信号通路可能参与了PV的发病。

     

    Abstract: Objective To investigate serum levels and mRNA expressions of tumor necrosis factor-like weak inducer of apoptosis (TWEAK) and its receptor CD163 from the patients with psoriasis vulgaris (PV). Methods Peripheral blood samples were obtained from 28 patients with PV and 15 healthy control subjects. Serum levels of TWEAK and CD163 were detected by ELISA, mRNA expressions of TWEAK and CD163 in peripheral blood were analyzed by real time-PCR, and protein expressions of TWEAK and CD163 were determined by flow cytometry. Results All the 28 PV patients were in progressive stage at the beginning of this study, 10 patients then recovered in convalescent stage after treatment. Compared to healthy controls, PV patients had higher serum TWEAK levels and lower serum CD163 levels. Serum TWEAK level in progressive stage was significantly higher than that in convalescent stage. Serum CD163 level were elevated significantly in convalescent stage compared with those in progressive stage. TWEAK mRNA expression in PV patients were significantly lower than that in healthy controls, but there was no significant differences of CD163 mRNA expression. TWEAK expression in monocytes in progressive stage and convalescent stage were significantly higher than that of controls, CD163 expression in monocytes in progressive stage and convalescent stage significantly lower than that in controls. No correlations wene found between psoriasis area and severity index (PASI) score and expression of TWEAK and CD163. Conclusion TWEAK/CD163 pathway may play a role in PV.
