p57kip2 基因在子痫前期患者胎盘表达的研究

Expression of p57kip2 Gene in Placenta Tissue of Patients with Pre-eclampsia

  • 摘要: 目的 探讨p57 kip2基因的核酸及蛋白水平在正常孕妇和子痫前期患者胎盘上的表达,探索p57 kip2基因与子痫前期发病之间的关系。方法 运用免疫组织化学SP法对p57 kip2基因在胎盘组织中的表达进行定位检测;通过逆转录聚合酶链反应法(RT-PCR)对37例重度子痫前期、22例轻度子痫前期及53例正常妊娠胎盘组织中的p57 kip2基因的核酸水平进行检测;通过免疫印迹法对纳入对象的p57 kip2蛋白表达水平进行检测。结果 免疫组化结果显示p57 kip2基因选择性表达于胎盘滋养细胞的细胞核中。RT-PCR结果显示:与正常妊娠组比较,轻、重度子痫前期组p57 kip2基因mRNA表达无明显变化(P>0.05),重度子痫前期组p57 kip2基因的mRNA表达水平低于轻度子痫前期组〔P<0.05,95%CI:(-0.736 9,-0.095 8)〕。免疫印迹结果显示:与正常妊娠组比较,轻、重度子痫前期组p57 kip2蛋白表达无明显变化(P>0.05),但重度子痫前期组p57 kip2蛋白的表达水平低于轻度子痫前期组〔P<0.05,95%CI:(-1.441 7,-0.162 4)〕。结论 p57 kip2基因的核酸和蛋白在重度子痫前期患者胎盘中的表达水平均低于轻度子痫前期患者,提示p57 kip2基因可能参与子痫前期的病理过程。

     

    Abstract: Objective To investigate the difference of p57kip2 gene expression in placenta tissue between normal pregnancy and pre-eclampsia. Methods The placenta tissues were collected from 37 cases of severe pre-eclampsia, 22 cases of mild pre-eclampsia and 53 cases of normal pregnancy. The localization of p57kip2 protein in placenta was studied by immunohistochemistry SP method. The mRNA and protein expression levels of p57kip2 gene were detected by RT-PCR and Western blot respectively. Results Immunohistochemistry result showed that p57kip2 protein was selectively expressed in the nucleus of placental trophoblast cells. The mRNA and protein expression levels of p57kip2 in severe pre-eclampsia were significantly lower than those in mild pre-eclampsia 〔P <0.05,mRNA 95%CI :(-0.736 9,-0.095 8), protein 95%CI :(-1.441 7,-0.162 4)〕. Conclusion The expression of p57kip2 gene in placental tissue may be involved in the pathogenesis of pre-eclampsia.
