绿原酸对肝星状细胞增殖、细胞外基质生成及降解的影响

Effect of Chlorogenic Acid on Hepatic Stellate Cell Proliferation, Generation and Degradation of Extracelluar Matrix

  • 摘要: 目的 研究绿原酸(CGA)对肝星状细胞增殖、胶原蛋白(Collagen) Ⅰ、Collagen Ⅲ、基质金属蛋白酶抑制剂-1(TIMP-1)及基质金属蛋白酶-2(MMP-2)表达及分泌的影响。方法 体外培养大鼠肝星状细胞株HSC-T6,随机分为5组:阴性对照组、血小板衍生生长因子(PDGF)组、PDGF+ CGA (12.5 μg/mL)组、PDGF+ CGA (25 μg/mL)组和PDGF+CGA (50 μg/mL)组。干预24 h后,MTT法检测HSC-T6的增殖情况;RT-PCR检测Collagen Ⅰ、Collagen Ⅲ、TIMP-1及MMP-2 mRNA表达;ELISA检测细胞上清液中Collagen Ⅰ、Collagen Ⅲ、TIMP-1及MMP-2的蛋白含量。结果 CGA可明显抑制PDGF诱导的肝星状细胞增殖(P<0.05),且随着剂量的增加,抑制作用逐渐增强(P<0.05);CGA可明显抑制PDGF诱导的Collagen Ⅰ、CollagenⅢ、TIMP-1的表达和分泌(P<0.05),但对MMP-2的表达和分泌无明显影响。结论 CGA可通过抑制肝星状细胞增殖及细胞外基质合成、促进细胞外基质降解来发挥抗肝纤维化作用。

     

    Abstract: Objective To investigate the effects of chlorogenic acid (CGA) on hepatic stellate cell proliferation and the expression and secretion of Collagen Ⅰ, Collagen Ⅲ, tissue inhibitors of metalloproteinase-1 (TIMP-1) and matrix metalloproteinase-2 (MMP-2). Methods An immortalized rat hepatic stellate cell (HSC) line was cultured In vitro. The cells were divided into 5 groups: control group; platelet-derived growth factor (PDGF) (10 ng/mL), PDGF+CGA (12.5 μg/mL), PDGF+CGA (25 μg/mL), PDGF+CGA (50 μg/mL) and CGA (50 μg/mL) group. After 24 hours treatment, the proliferation of HSC was detected by MTT method. The mRNA expression of Collagen Ⅰ, Collagen Ⅲ, TIMP-1 and MMP-2 were detected by RT-PCR. The protein levels of Collagen Ⅰ, Collagen Ⅲ, TIMP-1 and MMP-2 in the culture supernatant of HSC were measured by ELISA. Results PDGF increased the hepatic stellate cell proliferation, the mRNA expression and the protein levels of Collagen Ⅰ, Collagen Ⅲ and TIMP-1 (P<0.05), which were significantly decreased by CGA (P<0.05). However, CGA had no significant influence on the expression of MMP-2. Conclusion The antifibrotic effect of CGA may be related with the inhibition of hepatic stellate cell proliferation and generation of extracelluar matrix and promotion of extracelluar matrix degradation.
